FDA Approves Yuviwel for Children With Achondroplasia

What Was Approved?

On February 27, 2026, the U.S. Food and Drug Administration approved Yuviwel (navepegritide) to increase linear growth in pediatric patients 2 years of age and older with achondroplasia and open epiphyses.

The approval was granted through the FDA’s accelerated approval pathway.

What Is Achondroplasia?

Achondroplasia is a genetic condition that affects bone growth and is the most common form of disproportionate short stature.

It is associated with overactivity of the FGFR3 signaling pathway, which interferes with normal growth of long bones.

How Does Yuviwel Work?

Yuviwel is a C-type natriuretic peptide, or CNP, analog.

It is designed to promote bone growth by counteracting signaling pathways that restrict growth in achondroplasia.

How Is Yuviwel Given?

Yuviwel is given by subcutaneous injection once weekly.

The dose is based on body weight and should be adjusted as the child grows. Treatment should be discontinued when there is no further growth potential because the growth plates have closed.

What Did the Clinical Trial Show?

The pivotal trial enrolled 84 children with genetically confirmed achondroplasia.

After 52 weeks, children receiving Yuviwel grew an average of 1.5 centimeters more than those receiving placebo.

These results reflect the study population and do not guarantee the same amount of growth for every child.

Why Was It Granted Accelerated Approval?

The approval was based on improvement in annualized growth velocity, or AGV.

FDA states that increased growth velocity is reasonably likely to predict an increase in final adult height, but that final adult height benefit has not yet been confirmed.

A confirmatory study is ongoing.

What Are the Main Safety Concerns?

The FDA label warns about low blood pressure.

Symptoms may include:

  • Dizziness
  • Lightheadedness
  • Fainting
  • Blurred vision

The most common adverse reactions included vomiting, injection-site reactions, pain in the arms or legs, and nausea.

Bottom Line

Yuviwel (navepegritide) is a newly FDA-approved once-weekly injectable treatment for children age 2 and older with achondroplasia who still have open growth plates.

Because the approval was based on improved growth velocity rather than confirmed final adult height, long-term clinical benefit still needs to be verified in follow-up studies.
