What Was Approved?
On December 30, 2025, the U.S. Food and Drug Administration approved Nereus (tradipitant) for the prevention of vomiting induced by motion in adults.
Nereus is intended to be taken before an event expected to cause motion-related vomiting.
How Does Nereus Work?
Nereus is a substance P/neurokinin-1, or NK-1, receptor antagonist.
Blocking NK-1 receptors can interfere with signaling pathways involved in nausea and vomiting.
How Is Nereus Taken?
The recommended dose is either:
- 85 mg, or
- 170 mg
taken as a single oral dose approximately 60 minutes before an event expected to cause vomiting from motion.
It should be taken on an empty stomach, at least one hour before or two hours after a full meal.
Only one dose should be taken within a 24-hour period.
What Are the Common Side Effects?
The most common adverse reactions reported in clinical studies were:
- Sleepiness
- Headache
- Fatigue
Because Nereus can cause drowsiness or impair alertness, caution is important with driving or operating machinery.
Are Drug Interactions Important?
Yes.
Strong CYP3A4 inhibitors can increase tradipitant exposure and may increase the risk of adverse effects.
Other medicines that cause central nervous system depression may also increase impairment.
Patients should review their medications with a pharmacist or prescriber before use.
Is Long-Term Frequent Use Established?
Not fully.
The FDA label states that the safety of Nereus for prevention of motion-induced vomiting beyond 90 doses has not been established in clinical trials.
Bottom Line
Nereus (tradipitant) is an FDA-approved oral medication for preventing vomiting caused by motion in adults.
It is taken as a single dose before anticipated motion exposure, but it can cause sleepiness and has important drug-interaction considerations.
